Amino acid alignment of C. roseus ZCT proteins shows conservation of characteristic motifs. The B-box and L-box regions are thought to play a role in nuclear localization and protein–protein interaction, respectively. The Cys2-His2 zinc fingers (with the Cys and His residues highlighted in blue) contain the QALGGH motifs responsible for binding to the major groove of the target DNA. The EAR or LxLxL motif is a strong repression domain and is consistently present in most ZCT proteins. tZCT6 refers to the truncated version of the ZCT6 protein and contains one instead of the two Cys2-His2 zinc finger domains found in the other ZCTs (color figure online)
