GL7 Breg cells are expanded in mice with lupus-like disease. Experimental lupus induced by the hydrocarbon oil 2,6,10,14-tetramethylpentadecane (TMPD; also known as pristane) displays many key immunological and clinical features of human SLE. 0.5 ml 2,6,10,14-tetramethylpentadecane per mice were injected i.p. into IL-10 reporter mice. On day 1, 7, 14 and 21 after injection, lymphocytes were isolated from spleen, lymph node (LN) and peripheral blood mononuclear cells (PBMC) and analyzed by FACS. Quadrants (A, C) indicate percentage of IL-10-expressing (EGFP+) GL7 and GL7+ B cells. (B, D) Statistical analysis of the percentages of GL7IL-10+B and GL7+IL-10+B cells. Data represent at least three independent experiments and were analyzed by Student’s t-test (two tailed). Error bars, s.e.m. *P < 0.05, **P < 0.01.
