Macrophage knockdown of ORC6 protects mice from LPS-induced septic shock. The ORC6-mKD C57/BL6 mice, the aav-shC mice, and control mice (Ctrl) were injected intraperitoneally with LPS (50 mg/kg). Mice mortality (%) was recorded 24 h after LPS administration (A). Tail vein serum samples were collected 2 h after LPS stimulation, and IL-1β, TNFα, and IL-6 levels were determined by ELISA (B-D). Additionally, the murine peripheral blood mononuclear cells (PBMCs) were isolated 2 h after LPS stimulation. The mRNA and protein expression of ORC1 and ORC6 was examined (E and F), and NFκB pathway activation was assessed by p65 DNA-binding assay (G). Data were presented as mean ± standard deviation (SD, n = 5), with statistical significance indicated by *P < 0.05 compared to “Ctrl” group.
