Infectivity of TMV derivatives encoding ΔMP:GFP fusion proteins. TMV variants encoding either wild-type MP:GFP or mutant ΔMP:GFP derivatives were inoculated onto leaves of N. tabacum cv. Xanthi NN plants to test for local lesion development (A) in response to local virus movement. (B) Results obtained following inoculation of nontransgenic (NN) and MP-transgenic (NNMP) plants, showing the presence (+) and absence (--) of local lesion formation. The ability of a virus mutant to induce the formation of lesions on MP-transgenic but not nontransgenic plants indicates that the virus replicates but encodes a ΔMP:GFP fusion protein deficient in TMV RNA transport.
