The experiments were carried out as in Figure 4(A), except that, prior to stimulation with insulin for 3 min or EGF for 6 min, the cells were incubated for 1 h without (−) or with (+) 50 nM rapamycin, 2 μM PD 184352 or 100 nM wortmannin. The membranes were immunoblotted with the antibodies described in the legend to Figure 4, as well as with antibodies that recognize the active, phosphorylated forms of ERK1 and ERK2 and ribososomal protein S6 phosphorylated at Ser²³⁵ (pS6).
