Schematic procedure of micro-scale cultivation in 96-well plates. First, the plate is prepared (a) by adding 150 µL of agarose-containing medium to each well. Upon solidification of the media, inoculation (b) occurs with 50 µL of liquid pre-culture to every second column and leaving the outermost colums empty.
