PKCδ activation by PMA alone is sufficient to trigger CCR and caspase-3 cleavage in post-mitotic neurons. Primary cortical neurons were transfected with LacZ-shRNA or PKCδ-shRNA for 24 h, followed by exposure to 40 ng/mL PMA for an additional 8 h, before detection of cyclin D1 (B) or an additional 24 h before detection of PKCδ (A), PCNA (C), and p-Histone H3 (D), as well as pro- and cleaved caspase 3 (E). α-Tubulin served as the internal control for equal loading of proteins in each lane.
