Genital lymph node cells were collected from immunized mice 2.5 days after challenge, incubated in the presence of PMA, calcium ionophore, and brefeldin A, and stained for cell surface markers and intracellular IFN-γ. Cells gating positive for IFN-γ were considered activated. Gates were set to lymphocytes, and 3,500 to 10,000 events were counted. Representative results from one of two experiments are shown. Cells from unimmunized mice did not stain positive for IFN-γ (not shown).
