Type IV DGKs have different effects on Ras signaling. Immortalized fibroblasts from either wild-type, dgki–/– (A), or dgkz–/–(B) mouse embryos were transfected with the PathDetect plasmid constructs and starved overnight. They were then treated with platelet-derived growth factor (PDGF, 5.5 h, 40 ng/ml), and luciferase activity was detected.
