To approximate the effect of transcriptional silencing at the nuclear membrane, nuclear borders were traced using DNA labeling (¹³C-thymidine) as a guide. In parallel, nuclear borders were traced using the RNA labeling (¹⁵N-uridine) as a guide. The respective areas were extracted, and the % ¹⁵N-uridine unlabeled area at the nuclear margin was calculated by the following formula: area (% unlabeled) = 100 x (DNA area − RNA area)/DNA area.
