The block in intermediate gene expression is independent of DNA replication. BS-C-1 or L929 cells were transfected with p30/300 and subsequently infected with 5 PFU of WR or ts25 per cell in the presence of absence of AraC at the indicated temperature. Lysates were prepared at 24 h after infection and assayed for β-galactosidase activity (optical density at 495 nm [OD₄₉₅]).
