Subconjunctival ssAAV2-STC-1-FLAG lowers IOP in pigment dispersion and steroid-induced ocular hypertension mice. A, DBJ/2J mice (n = 14) were injected subconjunctivally with ssAAV2-STC-1-FLAG (2μL; 6 × 109 VG) in 1 eye and ssAAV2-GFP (2μL; 6 × 109 VG) in the fellow eye. Significant, sustained IOP lowering was seen in the eye injected with ssAAV2-STC-1-FLAG through week 12. Of note, ssAAV2-STC-1 blunted the naturally occurring rise in IOP in the DBA/2J model. B, Steroid-induced ocular hypertension was induced in 1 eye of C57BL/6J mice (n = 15). Mice were injected subconjunctivally in the ocular hypertension eye with ssAAV2-STC-1-FLAG (2μL; 6 × 109 VG, n = 8) or ssAAV2-GFP (2μL; 6 × 109 VG, n = 7). Significant sustained IOP lowering was seen in the eye injected with ssAAV2-STC-1-FLAG.
