Upshifts in the thermal unfolding curve of response regulator HsrA triggered by ligands IV and XII. The high-throughput screening (HTS) of the Maybridge HitFinderTM chemical library for HsrA ligands was assessed by a fluorescence-based thermal shift assay. Mixtures of DMSO (vehicle) instead of compounds were used as reference controls. Any compound that preferentially binds to the native state of HsrA would increase protein stability and cause an increase in the midpoint temperature of unfolding (Tm). Compounds that clearly increased the thermal stability of HsrA observed in the reference controls (Tm ≥ 5 °C) were considered as HsrA ligands.
