Workflow for subclonal reconstruction. Illustration of an oral potentially malignant disorder (OPMD) and oral squamous cell carcinoma (OSCC) lesions and normal buccal mucosa sampled from the same patient with both brush and tissue biopsies. The samples were aligned, and somatic mutations were called from the aligned reads. CNA profiles were constructed using ASCAT and SNV CCFs were then clustered to identify subclonal lineages in the samples and phylogenetic reconstruction was done.
