Virus production was assayed as RT activity in the supernatant of transfected cells. Values were converted to percentages of wild-type level for each of nine independent transfections and were averaged.

Viral infectivity was assayed by measuring RT activity in the supernatant of infected CEM cells.

Infectivity in the MAGI assay in a single round of infection. The numbers of blue cells were normalized to the RT activity of input virions and are reported as percentages of wild type.

Virions obtained from transfected 293T cells were used to infect CD4-positive HeLa indicator cells, which were then lysed. Viral DNA was amplified by PCR.
