PAR₂-mediated sensitisation of TRPV4 is not affected by BAPTA-AM. Oocytes were injected with cRNA coding for TRPV4 0.5 ng or TRPV4 0.5 ng/PAR₂ 10 ng and incubated for 2 days in ND96 solution. GSK1016790A-mediated whole-cell currents were measured using the two-electrode voltage-clamp technique. Oocytes were pre-incubated for 3h in Ca²⁺-free solution in the absence (−) or in the presence (+) of BAPTA-AM (100 μM). After a subsequent pre-incubation for 5 min in trypsin (8 nM), TRPV4 was activated by GSK1016790A (50 nM). Columns represent mean ΔI_GSK1016790A values. N indicates the number of batches of oocytes.
