(A) Total CD4s were pretreated with 100 μM HODHBt, 10 μM AC-484, and 5 μM AC-484 for 2 hours prior to the addition of IL-2. pSTAT5 levels were measured by flow cytometry 1, 24, and 48 hours after stimulation with IL-2 (n = 6). Wilcoxon matched-pairs signed-rank test was used to calculate P values (*P < 0.05; **P < 0.01). (B) Total CD4 T cells were pretreated with 100 μM HODHBt or 10 μM AC-484 for 2 hours prior to the addition of IL-15. pSTAT5 levels were measured by flow cytometry 48 hours after stimulation with IL-2 (n = 3).
