Stimulation of ABCA1 protein abundance and cholesterol efflux by Oxy210 in RAW264.7 macrophages. (A) Raw264.7 cells were grown to confluence in the DMEM containing 10% FCS and then incubated with Oxy210 (5 μM) or vehicle for 24 h in medium containing 0.1% FCS. Cells were then lysed, and ABCA1 and GAPDH in lysates were detected by Western blotting and visualized and quantitated using G:Box imaging system. Data from a representative experiment are reported (n = 6, biological replicates) as mean ± SD (** p < 0.01 vs. control). (B) Raw264.7 cells were grown to confluence and labeled with [3H] cholesterol for 48 h in a medium containing 10% FCS. Cells were then incubated for 24 h with Oxy210 (5 μM) or vehicle in a medium containing 0.1% FCS. Cells were then washed and incubated for 2 h in a serum-free medium containing purified human apoA-I (25 μg/mL) or without apoA-I as control. Specific cholesterol efflux is shown and was expressed as a percentage of labeled cholesterol moved from cells to apoA-I-containing media minus that in the absence of apoA-I (blank).
