(B and C) WT or D652A FLAG Pol η was immunoprecipitated from 293T cells co-expressing HA ubiquitin (B) or HA NEDD8 (C). The eluent and lysate were immunoblotted as indicated. Relative mono-UBLylation was calculated based on the ratio of higher to lower FLAG Pol η bands. Mono-ubiquitination and mono-NEDDylation was calculated based on the ratio of HA to FLAG bands. The graph represents calculated values from three independent experiments. Error bars represent standard deviation.
