Suboptimal NT concentrations were 0.2 nM NGF, 0.6 nM BDNF, and 0.2 nM NT-3 for TrkA-, B-, and C-expressing cell lines, respectively. Survival data was normalized relative to DMSO (0% survival) and optimal neurotrophin (2.0 nM NGF, 1.0 nM BDNF, 2.0 nM NT-3; 100% survival). EC50 values were calculated using the nonlinear regression “{agonist} vs response – Variable slope (four parameters)” analysis, and Kd’s were calculated by first subtracting nonspecific binding, then using the nonlinear regression “One site – specific binding” analysis in GraphPad Prism 10.2.
