Phosphate content of leaf starch

The phosphate content of starch from Arabidopsis leaves or maize endosperm tissue was measured in units of μmol/mg starch. Starch isolated from Arabidopsis leaves at the end of the light phase or from imbibed mature maize kernels, was dried, then suspended in buffer. Phosphate content measurements were conducted on equivalent amounts of starch in duplicate or triplicate, as indicated.
