Overview of the bacteriophage recombineering of electroporated DNA (BRED) technique. Phage and synthetic DNA are co-electroporated into a M. smegmatis mc2155 strain carrying a plasmid encoding homologous recombination proteins (gp60 and gp61). Homologous recombination between phage and synthetic DNA, with subsequent plating of transformed bacteria, allows the obtention and screening of plaques to identify deletion mutants [56].
