Antigen-specific memory T and B cell responses induced by the vaccination. Lung, spleen, and bone marrow cells were harvested at day 7 post-infection from the immunized and infected mice. Lung and spleen cells were labeled with CFSE and cultured for 5 days in the presence of sPR8 (10 µg/ml) stimulation. T cell proliferation was then determined via flow cytometry (a). Bone marrow cells and spleen cells were also harvested, then incubated with sPR8 (10 µg/ml) for 1 day and 7 days, respectively. Subsequently, antigen-specific IgG production was measured by ELISA (b). Means ± standard errors were shown with individual dots. One-way ANOVA and Tukey’s multiple comparison test were performed for statistical analysis. *p < 0.0332; **p < 0.0021; ***p < 0.0002; ****p < 0.0001 compared between the indicated groups.
