(B) Southern blot of DNA isolated from P3HR1 cells electroporated with the indicated plasmids. After cleavage with BamHI, the electrophoretically separated DNA fragments were transferred to a nitrocellulose membrane and probed with a 32P-labeled EcoRI-Dhet probe. As indicated by the increase in the amount of the BamHI-Nhet fragment, linear EBV genomes were efficiently amplified only in wild-type Zta-transfected cells. Lanes 1 to 5, DNA isolated from electroporated P3HR1 cells; lane 6, cosmid 302–21 DNA digested with BamHI.
