In the absence of Wnt9a, Fzd9b is ubiquitinated at K437, leading to its lysosomal degradation. Wnt target gene activation is shut off. When Wnt9a is present, the Fzd9b receptor complex, including LRP5/6 is assembled, leading to β-catenin translocation to the nucleus, and activation of Wnt9a/Fzd9b target genes that drive hematopoietic stem cell proliferation.
