The combination of Panobinostat and Doxorubicin increases the expression of DNA damage and apoptosis markers and decreases the expression of DNA damage response proteins. (A,B) Ewing sarcoma patient cells were treated with Panobinostat (50 nM) alone or in combination with Doxorubicin (125 nM) for 24 h and protein expression was determined by Western blotting with the indicated antibodies (A). Representative uncropped blots are shown in Figure S4. Signal intensities were quantified using the AzureSpot software to select bands and subtract background. Intensities were normalized to ß-Actin, and fold changes were calculated for each drug-treated sample over vehicle-treated ones. One-way ANOVA with Dunnett’s multiple comparisons test indicates significance: ns: no significance, * p < 0.05, ** p < 0.01, *** p < 0.001, and **** p < 0.0001 (B).
