Schematic illustration how XBP1 facilitates NF-κB-p65 nuclear translocation via regulating MT2 transcription in macrophages during liver IRI. XBP1 is increased in macrophages of the IR-stressed liver, and then is bound to MT2 promoter to inhibit its transcription. However, MT2 enhances IKBα transcription and represses NF-κB-p65 phosphorylation and nuclear translocation. Thus, XBP1 facilitates NF-κB-p65 nuclear translocation and contributes to macrophage-originated sterile inflammation in the IR liver.
