IGF2BP1-3 do not possess redundant or overlapping functions on the CDC5L protein in 1q+ MM. A. IG2BP1 and IGF2BP2 both exhibits significantly higher expression in 1q+ MM patients from the MMRF CoMMpass dataset. B. IGF2BP2 does not possess prognostic implications for progression-free survival (PFS) (p = 0.097) and overall survival (OS) (p = 0.66) in 1q+ MM patients. C. A notable reduction in IGF2BP3 mRNA expression level (p = 0.016) was observed in IGF2BP1-KD NCI-H929 cells, while IGF2BP2 exhibited minimal alteration. The IGF2BP1 (p = 0.033) and IG2BP2 (p = 0.003) mRNA expression level both increased significantly in IGF2BP3-KD NCI-H929 cells. D. The reduction in IGF2BP3 protein level has a marginal effect the increase of IG2BP1 protein level (p = 0.055). E. The reduction in IGF2BP3 protein level does not have any discernible impact on the protein abundance of CDC5L.
