Western blot was used to detect the uptake of amyloid Aβ1-42 into cells. The results demonstrated that Aβ1-42 protein levels in NC-HEK293T cells, OATP1A2-HEK293T cells, and human astrocytes increased gradually with increasing time (24–72 h) and Aβ1-42 concentration (0.4–2.5 μM). This suggests that the uptake of Aβ1-42 by these cells is dependent on both time and concentration. Notably, the uptake of Aβ1-42 in OATP1A2-HEK293T cells was significantly higher than that in the NC-HEK293T control cells, especially at 48 h and 72 h of culture. At these time points, the uptake of Aβ1-42 in OATP1A2-HEK293T cells was nearly twice as high as that in the NC-HEK293T group. Additionally, HNA (human astrocytes) were included in the study.
