Functional analysis of MDR1NAM. MDR1NAM and MDR1 were transiently expressed in HeLa cells. The cell surface expression of each protein was compared by MRK-16 labeling followed by FACS analysis. The transport activity of transiently expressed MDR1NAM was analyzed by fluorescent substrate accumulation assay at a final concentration of 1 μM bisantrene, 0.5 μM rhodamine-123, or 3 μM daunomycin. A zoom-in version of the FACS result is used for the bisantrene panel.
