MAGEL2 acts as an E3 ubiquitin ligase enhancer and interacts with the E3 RING ubiquitin ligase TRIM27, and the deubiquitinating enzyme USP7, which buffers ubiquitination levels of substrates such as the WASH complex. 32 , 55 WASH ubiquitination by the MUST complex has been shown to be involved in retromer recycling. 56 The figure highlights several neurodevelopmental disorders linked to variants in genes encoding MUST proteins or their interaction partners.
