Overexpressing MFN2 can reverse IH-EXOS-mediated pyroptosis of HT22 cells induced by miR-20a-5p (n=3). Following co-transfection of miR-20a-5p mimics and MFN2 silencing or overexpression with or without IH-EXOS exposure, cells were analyzed via (A) qRT-PCR was used to detect the expression of miR-20a-5p. (B and C) Western blot and qRT-PCR were used to detect the expression of MFN2, GSDMD, and Caspase-1 protein and mRNA. (D) ELISA was used to detect IL-1β, IL-6, IL-18, and TNF-α levels. Statistical analysis of the data was performed using two-way ANOVA followed by Bonferroni’s multiple comparison tests, df (5, 12). *P<0.05.
