Immunoblot analysis with selected phage-displayed peptides. OMPs isolated from NTHi strain R2866 were separated by SDS-PAGE and blotted onto PVDF membranes (Millipore). The membranes were probed separately with unselected f88-4/15-mer library and phage clones hi3/17, hi3/2, hi3/12, and hi3/13, respectively. Bound phage-displayed peptides were detected by using mouse anti-M13 antibody (Amersham Pharmacia Biotech) and then by using rabbit anti-mouse immunoglobulin G conjugated to horseradish peroxidase (Sigma). The bands on the immunoblot were visualized by using the TMB membrane peroxidase substrate system (Kirkegaard & Perry Laboratories). SDS-PAGE broad-range prestained markers were used (Bio-Rad).
