Reduction of mTOR protein expression by siRNA specific to mTOR increases HCV replication. Replicon cells were transfected with mTOR siRNA (+) or nontargeting siRNA (−). (A) After 30 h, cells were lysed and mTOR, NS5A, and actin were detected by Western blotting. (B) Total RNA was isolated and cDNA was synthesized by use of HCV minus RNA strand-specific primer. Real-time PCR analysis was performed with HCV 5′ non-translated region-specific primers. Average values from three independent experiments ± standard deviations are shown.
