Bud2p provides a landmark for yeast and hyphal daughter cell site selection. Stationary-phase C. albicans strains JB6284 (WT), CA8880 (rsr1Δ/Δ), and DH4453 (bud2Δ/Δ) were subcultured under YF growth conditions for 4 h and stained with calcofluor white to visualize bud scars (top) or grown under HF conditions (bottom) and analyzed by DIC microscopy for branch position (asterisks) relative to hyphal septa (arrows). Scale bars, 5 and 10 μm (upper and lower panels, respectively).
