Wild-type Smad3 and the Smad3 (143–248) mutant are functional in yeast cells. (A) Schematic representation of the yeast expression vectors that were utilized in the assays of B–D. (BD) Functional assays which established that GAL4-Smad3 (wt) or GAL4-Smad3 (143–248) mutant are functional in yeast cells. The assays were based on the ability of transformed GAL4-Smad4 (wt) or GAL4-Smad3 (143–248) to activate chromatin incorporated reporters consisting of 5 GAL4 binding sites linked to the β-galactosidase gene (B), the ADE1 gene (C) and the HIS3 gene (D). These assays were performed as described in Materials and Methods.
