Sporulation in NC1 and NC2 domain expression strains. Representative culminants from strains Ax3 (parental), ANC1, and ANC2 are shown in the panels on the left. Ax3 forms a normal fruiting body with a sorus, consisting of spores, perched on top of the cellular stalk. The unusual morphology of strain ANC2 varied in frequency from 50 to 90%, with the remaining culminants having a more normal appearance as shown in panel A. Spores were found only in the lower sorus when present. Panels on the right show images from the same field of view of a representative squash preparation of a single sorus. The field was imaged using either phase contrast (middle panels) or epifluorescence to reveal labeling with Calcofluor White ST (right panels) to show cellulose. Note that most ANC1 cells were Calcofluor negative and tended to lyse, that both ANC1 and ANC2 Calcofluor-positive cells tended to be less elongate than parental Ax3 cells (Table 3), and that ANC2 spores did not disperse as well under the coverslip owing to abnormal self-cohesiveness.
