Characterization of culB. (A) culB genomic locus. pBsr1 is a 4.1-kb plasmid with the selectable Bsr cassette that was inserted into the BamHI site of the culB gene (hatched rectangle). Genomic DNA from wild-type, culB (clones 1 and 2) or culB pkaR (clone 3) cells was digested with BclI, separated by agarose gel electrophoresis, blotted, and probed with the XbaI-BamHI genomic DNA fragment (hatched bar) in a standard Southern analysis.
