Membranes of A7r5 cells were fractionated on sucrose gradients (see the Experimental section) and fractions of 250 μl were collected. (a) Sucrose concentration (●) and protein content (○) of each fraction. (b) Representative Western blots for samples of 13 μl from the indicated fractions. The Mr of each band is shown calculated from the migration of standard Mr markers (MagicMarker-XP, Invitrogen). The 5-HT2A antiserum identified two similarly intense bands with Mr ∼55 and ∼65; the lower band was used for quantification. (c) Results from three independent experiments (means±S.E.M) are summarized with the intensity of caveolin 1 (■), V1A receptor (●), 5-HT2A receptor (○) and β-adaptin (□) bands each expressed as a percentage of the sum of the intensities of the bands across the entire gel.
