Scheme 1. Proposed schema for IFNγ-induced MEKK4 pathway.

Scheme 1

IFNγ activation of the IFNGR in keratinocytes results in activation of STAT1 via JAK2 phosphorylation and leads to antiviral defence response (left-hand side). IFNγ activation also leads to a flux of free calcium. Annexin II binding calcium contributes to Pyk2's calcium-dependent activation. MEKK4 is tyrosine-phosphorylated by Pyk2 and activates MKK6, which activates p38 MAPK and results in activation of transcription factors for prostaglandin responses (right-hand side).
