Phospholipid composition of mitochondrial nucleoids as a function of their isolation condition. Phospholipids extracted from three mitochondrial-nucleoid samples, each obtained from 25 mg protein equivalent of mitochondria at a NP-40 concentration of 0.25, 0.5 and 1.0% were loaded in lanes 2, 3 and 4, respectively, of a thin-layer chromatography chromatogram. A phospholipid sample extracted from 0.5 mg protein equivalent mitochondria was loaded in lane 5 as a control. A mixture of phospholipid markers, cardiolipin (CL, 20 μg), phosphoglycerol (PG, 10 μg), phosphatidylethanolamine (PE, 10 μg), phosphatidylinositol (PI, 20 μg) and phosphatidylcholine (PC, 10 μg) were loaded in lanes 1 and 6. The chromatogram was developed and stained as described in Materials and Methods.
