Immunoblotting analysis of mitochondrial-nucleoid proteins as a function of their isolation condition. Five mitochondrial-nucleoid samples prepared at a NP-40 concentration of 0, 0.25, 0.5, 0.75 and 1%, were loaded in lanes 2, 3, 4, 5 and 6, respectively, for SDS–PAGE. Each nucleoids sample loaded was obtained from 50 μg protein equivalent of mitochondria. A total of 0.5 μg protein equivalent of lysed mitochondria was loaded in lane 1 as a control. The following antibodies were used for the ensuing immunoblots: anti-α subunit of ATPase, anti-COXII, anti-porin and anti-actin.
