In organello mitochondrial DNA synthesis with different pulse and chase periods. Mitochondrial DNA synthesis was carried out as described in Materials and Methods. At 0.5, 1, 2, 4 and 8 h pulse period, respectively, an aliquot was taken from the reaction mixture and mitochondria (equivalent to 0.5 mg protein) were recovered by centrifugation, and subjected to PFGE and autoradiographic analysis. (A) PFGE autoradiogram of newly synthesized mtDNA at different pulse periods. Lanes 1–5 represent 0.5, 1, 2, 4 and 8 h pulse period, respectively. (B) PFGE-autoradiogram of newly synthesized mtDNA after different chase periods. Lanes 1–5 represent 0, 10, 30 min, 2 and 4 h chase periods, respectively, after an 1 h pulse. (C) The effect of ATA on mitochondrial DNA synthesis. This experiment is the same as that shown in (A), except 100 μM ATA was added to the reaction mixture 1 h after the start of the DNA synthesis reaction. Lanes 1–5 present samples taken at 1, 2, 4, 6 and 8 h, respectively, following the addition of ATA.
