tSNAPc is required for SL RNA gene transcription. (A) Western blot analysis of preimmune-depleted (PreIm) and anti-tSNAP42-depleted (α-tSNAP42) nuclear extracts. Only a portion of TBP was depleted from the extracts. (B) Transcription activity was lost in the tSNAPc-depleted extract (compare lanes 3, 4, and 9 with 1, 2, and 8). Depleted extracts were reactivated when supplemented with 5, 10, and 20 ng of affinity-purified tSNAPc (lanes 5 to 7) or 3, 6, and 12 ng of sucrose velocity-purified tSNAPc (lanes 10 to 12).
