(A) Coimmunoprecipitation (IP) experiments in transiently transfected C2C12 cells demonstrate in vivo interaction between FoxO1a and a constitutively active form of cGKIα (cGKI-CA, where the cGMP regulatory domain has been deleted). Western blotting (WB) was performed with an anti-FLAG antibody. (B) Reverse coimmunoprecipitation experiments in transiently transfected C2C12 cells demonstrate the in vivo interaction between endogenous cGKIα and a FLAG-tagged FoxO1a. C2C12 cells were induced to differentiate for 3 days. Western blotting was performed with an anti-cGKI antibody.
