Dipyridamole reversibility in Krebs-2 assays. Krebs-2 lysates programmed with mengovirus virion RNA were incubated for 4 h at 32°C in the presence (lanes 2 and 3) or absence (lanes 1 and 4) of 80 μM dipyridamole (DPM). The replication complexes were collected by centrifugation at 16,000 × g and then resuspended in fresh lysates in the presence (lanes 2 and 4) or absence (lanes 1 and 3) of 80 μM dipyridamole. [³²P]CTP was added and incubation was continued for 1 h. The samples were fractionated on 1% native agarose gels, and the labeled bands were visualized by autoradiography.
