Microscopy has long been a fundamental tool for detecting parasites in biological samples. While molecular diagnostic and immunological techniques have advanced, microscopy continues to play a vital role, particularly in resource-limited settings. The key advantages include its ability to directly observe parasites and their developmental stages, with their morphology often providing specific or suggestive identification; cost-effectiveness, rapidity, broad applicability, ability to provide additional diagnostic clues, requirement of minimal equipment, and applicability as a point of care test. However, there are certain limitations, for example, the accuracy of observations is heavily dependent on the skill of the observer and the inexperienced or inadequately trained personnel may overlook important diagnostic signs, a low parasite load may lead to underdiagnosis and artifacts which can potentially lead to false positives and is labor intensive and time-consuming.
Advances in technology have led to some Innovations in microscopy also. The integration of digital imaging, combined with artificial intelligence (AI)-based algorithms, enhances the accuracy of microscopy, making it more feasible in field settings. Moreover, advances in mobile cameras and AI-enabled tablets are making mobile-based microscopy increasingly viable, whereas lightweight microscopes allow for use in remote or field locations. The availability of rapid staining methods, including acridine orange, fluorophores, and molecular dyes, is improving the sensitivity of parasite detection. Thus, revisiting microscopy involves integrating modern technologies to address its limitations. In addition, enhancing the skills of personnel to preserve the expertise of microscopy remains crucial.
The January–June 2025 issue of Tropical Parasitology includes an opinion on revisiting Koch’s postulates and tailoring these for parasites. The original work on the development of novel antibody detection test for strongyloides, immunological biomarkers for leishmaniasis, rhinosporidiosis, and malaria epidemiology is included in this issue. A review article on helminths and immunomodulation, a face-to face interview with Dr. Abhijit Chaudhary are other features of this issue.
