Incorporation of gE mutants 277, 291, and 348-4 into extracellular virions. R-970 cells were infected with F-BAC, gE-null mutant F-gEβ, or gE ET domain mutant 277, 291, or 348-4 at 10 PFU/cell. The cells were labeled with [³⁵S]methionine from 6 to 18 h postinfection, cell culture supernatants were harvested and centrifuged at low speed, and then virions were partially purified on 20%/50% sucrose step gradients. Virions were pelleted and disrupted in NP-40-DOC buffer and centrifuged at high speed to remove insoluble debris, gE/gI was immunoprecipitated using a mixture of MAb 3114 and 3063 or gD immunoprecipitated using MAb DL6, and the proteins were subjected to electrophoresis using polyacrylamide gels.
