Control Groups Setup for Nuclear Off-targets. A) Setup of control groups for nuclear off-targets. The ‘c1’ group controlled for transfection effect. The ‘c2’ group controlled for nuclear genome off-targets, including both TALE-Array sequence (TAS)-associated off-targets and TAS-independent off-targets. Group ‘n’ was introduced to the MT-ND5 mutations. B) Demonstration of the c2 group in controlling TAS-dependent and TAS-independent off-targets. Control group c2 was transfected with two confronting DddAtox splits fused with TAS, rendering them non-functional in the mitochondrial genome. Nonetheless, the appropriate paring TAS-free DddAtox splits were also introduced in c2 group, facilitating an accurate representation of TAS-dependent and TAS-independent nuclear off-target effects that might manifest in the treatment group (group n).48
