Flowcytometric analysis for a preculture MNCs sample: The first histogram showed viability testing using 7 – AAD. 99.4 % of cells were negative for 7 – AAD (viable cells). The second panel (first dot plot) (CD45 FITC against Side scattering) showed that 44.8% of viable cells are CD45+ positive. The second dot plot (CD45 FITC against CD34 PE) showed that 5.4 % of cells are double positive for CD45 and CD34. The third dot plot (CD45 FITC against side scattering showed dim CD45+ CD34+ cells, which are 57.2% of double positive for CD45 and CD34. The final cell selection was CD34+/ CD45+ dim cells (Hematopoietic stem and progenitor cells).
